How TriC Folds Tricky Proteins
How chaperonins orchestrate the successful folding of even the most elaborate of proteins is a question of central importance. In two recent studies in Cell by Joachimiak et al. and Freund et al., a new class of TRiC substrate is identified, and how the chaperonin exploits its different subunits to extend its substrate repertoire and direct productive folding is revealed.
متن کاملF-like type IV secretion systems encode proteins with thioredoxin folds that are putative DsbC homologues.
F and R27 are conjugative plasmids of enteric bacteria belonging to the IncF and IncHI1 plasmid incompatibility groups, respectively. Based on sequence analysis, two genes of the F transfer region, traF and trbB, and three genes of the R27 transfer region, trhF, dsbC, and htdT, are predicted to encode periplasmic proteins containing a C-terminal thioredoxin fold. The C-X-X-C active-site motif o...
متن کاملCaspase Cleavage Motifs of Influenza Subtypes Proteins: Alternations May Switch Viral Pathogenicity
Background and Aims: The caspases are unique proteases that mediate the host cell apoptosis during viral infection. In this study, we identified the caspase cleavage motifs of H5N1 and H9N2 influenza viruses isolated during 1998-2012. Materials and Methods: Amino acid sequences of the eleven proteins encoded by the viruses as the caspase substrates downloaded from NCBI. The caspase cleavage mot...
متن کاملA parallel beta-sheet model system that folds in water.
Proteins display a vast array of tertiary structures, but only a few regular motifs are common at the level of secondary structure. Establishing sequence-stability relationships for the common secondary structures should enhance our understanding of proteinfolding preferences. Many groups have used model systems to examine R-helix1 and, more recently, antiparallel â-sheet2 conformational stabil...
متن کاملA model for extradenticle function as a switch that changes HOX proteins from repressors to activators.
The Drosophila EXD protein and its mammalian counterparts, the PBX proteins, have been proposed to function in HOX target selectivity. Here we show that exd function is required for the autoactivation phase of Dfd expression in the posterior head. Mutations that change the affinity of a small autoactivation element for EXD protein result in corresponding changes in the element's embryonic activ...
